In deze rol fungeer je als stevige gesprekspartner voor projectleiders en afdelingshoofden en adviseer je over de financi\u00eble sturing en beheersing van onderzoeksprojecten. Hieronder vallen perifere neuropathie, die gevoelloosheid of pijn in handen en voeten veroorzaakt, evenals cognitieve problemen die vaak worden aangeduid als \u201cchemobrain\u201d, wat het geheugen kan be\u00efnvloeden. Deze complicaties kunnen nog lang na afloop van de behandeling aanhouden, met een grote impact op de kwaliteit van leven en in sommige gevallen zelfs leidend tot dosisverlaging of het stopzetten van de therapie. Momenteel bestaan er geen effectieve behandelingen om deze ernstige bijwerkingen te voorkomen.<\/p>\n<p>In dit door KWF gefinancierde project onderzoeken we hoe door chemotherapie veroorzaakte DNA-schade de gentranscriptie in zenuwcellen verstoort en hoe dit bijdraagt aan neurotoxiciteit. Dit project is een nauwe samenwerking tussen de onderzoeksgroepen van Jurgen Marteijn, Debbie van den Berg en Femke de Vrij, waarin expertise op het gebied van DNA-schade en transcriptiestress wordt gecombineerd met geavanceerde modellen voor humane neuronale differentiatie. Met behulp van state-of-the-art neuronale differentiatiemodellen, live-cell imaging, DNA-reparatie- en functionele neuronale assays en genoomwijde CRISPR-screening willen we de moleculaire mechanismen achter chemotherapie-ge\u00efnduceerde neurotoxiciteit ontrafelen en nieuwe aangrijpingspunten voor therapeutische interventie identificeren. Deze complicaties kunnen nog lang na afloop van de behandeling aanhouden, met een grote impact op de kwaliteit van leven en in sommige gevallen zelfs leidend tot dosisverlaging of het stopzetten van de therapie. Momenteel bestaan er geen effectieve behandelingen om deze ernstige bijwerkingen te voorkomen.<\/p>\n<p>In dit door KWF gefinancierde project onderzoeken we hoe door chemotherapie veroorzaakte DNA-schade de gentranscriptie in zenuwcellen verstoort en hoe dit bijdraagt aan neurotoxiciteit. Dit project is een nauwe samenwerking tussen de onderzoeksgroepen van Jurgen Marteijn, Debbie van den Berg en Femke de Vrij, waarin expertise op het gebied van DNA-schade en transcriptiestress wordt gecombineerd met geavanceerde modellen voor humane neuronale differentiatie. Met behulp van state-of-the-art neuronale differentiatiemodellen, live-cell imaging, DNA-reparatie- en functionele neuronale assays en genoomwijde CRISPR-screening willen we de moleculaire mechanismen achter chemotherapie-ge\u00efnduceerde neurotoxiciteit ontrafelen en nieuwe aangrijpingspunten voor therapeutische interventie identificeren. These include peripheral neuropathy, causing numbness or pain in hands and feet, as well as cognitive problems often referred to as \u201cchemobrain\u201d, which can affect memory. These complications can persist long after treatment has ended, severely affecting quality of life and, in some cases, forcing dose reduction or therapy stop. Currently, there are no effective treatments to prevent these severe side effects.<\/p>\n<p>In this KWF-funded project, we investigate how chemotherapy-induced DNA damage disrupts gene transcription in nerve cells and how this contributes to neurotoxicity. This project is a close collaboration between the labs of Jurgen Marteijn, Debbie van den Berg and Femke de Vrij, combining expertise in DNA damage and transcription stress with advanced human neuronal differentiation models. Using state-of-the-art neuronal differentiation models, live-cell imaging, DNA repair and functional neuronal assays, and genome-wide CRISPR screening, we aim to uncover the molecular mechanisms underlying chemotherapy-induced neurotoxicity and identify novel targets for therapeutic intervention.
