You will focus on analysing single cell long read RNAseq data generated by our team. You will work to discover cell specific isoforms and relate them to pathological signatures and genetic risk factors in ALS patient tissue. You will apply supervised and unsupervised machine learning methods using a local high performance computing cluster. You will work closely with experts in ALS, bioinformatics, neuroscience and RNA biology. You will support in the supervision of more junior researchers and students.

Als farmaceutisch medewerker verricht je diverse logistieke handelingen. Denk aan het ontvangen, verzamelen en verzenden van (patiënt specifieke) geneesmiddelen in de ziekenhuisapotheek. Ook beheer je de afdelingsvoorraad van de (verpleeg)afdelingen en satellietapotheken en houd hier toezicht op. Samen met jouw collega’s streef je naar een optimale beschikbaarheid van geneesmiddelen voor de patiënten.

De werktijden zijn variabel; doordeweeks werk je tussen 07.30 en 17.30 uur en in het weekend en feestdagen tussen 08.00 en 14:00 uur. De wisselende werktijden en de daarbij behorende pauzes is afhankelijk van de ingeroosterde diensten.

Research Technician — Lysosome Biology in Zebrafish (CLN3/Batten Disease)

In this translational project, you will apply recently developed zebrafish Lyso-IP/-Tag lines to interrogate intra-lysosomal proteomes, lipidomes and metabolomes in cln3-deficient models. The goals are to (i) uncover lysosomal pathways perturbed by CLN3 loss using LC-MS-based proteomics and metabolomics of enriched lysosomes from larvae and adult tissues (eye, brain), and (ii) test how different energy sources (lipids, glucose, amino acids) influence disease biomarkers (e.g., GPDs) and lysosomal morphology in vivo. You will work closely with Dr Peter van Hasselt (paediatrician for metabolic diseases), core facilities (Proteomics, Lipidomics, Metabolomics), and collaborators at the Hubrecht Institute.

Key responsibilities

• Maintain and manage zebrafish lines: breeding, husbandry, genotyping, and line crosses.
• Prepare lysosome-enriched fractions via Lyso-IP from larvae and adult tissues; perform QC by Western blot and basic fraction purity checks.
• Coordinate and execute sample preparation for LC-MS proteomics, lipidomics and metabolomics; liaise with core facilities and manage sample metadata.
• Perform fluorescence/confocal imaging of fluorescently tagged lysosomes; quantify lysosome number/size and morphology across conditions.
• Microinjection experiments of zebrafish embyos and phenotypic characterization; assess survival, morphology, locomotor behavior, and downstream biomarker changes.
• Maintain accurate lab records, SOPs, and inventory; ensure biosafety and animal welfare compliance; contribute to data organization and basic analysis.
• Collaborate within a multidisciplinary team and communicate results in lab meetings.

Samen met je collega’s zorg jij ervoor dat ons LIS niet alleen actueel is, maar ook toekomstbestendig blijft. Jij werkt met GLIMS en combineert beheer met innovatie om laboratoriumprocessen optimaal te laten verlopen.

• Je optimaliseert de LIS-inrichting, koppelingen en werkprocessen.
• Je ondersteunt collega’s in vakgebieden zoals microbiologie, virologie, immunologie, bloedbank, klinische chemie en apotheek.
• Je richt studies in, sluit apparaten aan en onderhoudt de koppelingen met het EPD (HiX).
• Storingen los je snel op en je denkt in oplossingen.
  Je draait mee in bereikbaarheidsdiensten en zorgt voor heldere rapportages.
• Je test verbeteringen zodat ons laboratorium 24/7 blijft presteren op het hoogste niveau.
